工作原理
凱(kai)氏(shi)(shi)定(ding)(ding)氮(dan)(dan)法(fa)是(shi)測(ce)定(ding)(ding)化(hua)合(he)物(wu)或混合(he)物(wu)中總(zong)氮(dan)(dan)量(liang)(liang)(liang)的(de)一種(zhong)方法(fa)。即在有(you)催化(hua)劑的(de)條件下(xia),用濃硫酸(suan)消(xiao)化(hua)樣品將有(you)機氮(dan)(dan)都轉變成無(wu)機銨(an)鹽(yan),然后(hou)在堿性條件下(xia)將銨(an)鹽(yan)轉化(hua)為(wei)氨,隨水蒸(zheng)氣蒸(zheng)餾出來并為(wei)過量(liang)(liang)(liang)的(de)硼酸(suan)液吸收,再以標(biao)準鹽(yan)酸(suan)滴定(ding)(ding),就(jiu)可(ke)計算(suan)出樣品中的(de)氮(dan)(dan)量(liang)(liang)(liang)。由于蛋白(bai)質含(han)(han)(han)氮(dan)(dan)量(liang)(liang)(liang)比較恒定(ding)(ding),可(ke)由其(qi)氮(dan)(dan)量(liang)(liang)(liang)計算(suan)蛋白(bai)質含(han)(han)(han)量(liang)(liang)(liang),故(gu)此法(fa)是(shi)經典的(de)蛋白(bai)質定(ding)(ding)量(liang)(liang)(liang)方法(fa)。因其(qi)蛋白(bai)質含(han)(han)(han)量(liang)(liang)(liang)測(ce)量(liang)(liang)(liang)計算(suan)的(de)方法(fa)叫做凱(kai)氏(shi)(shi)定(ding)(ding)氮(dan)(dan)法(fa),故(gu)被稱為(wei)凱(kai)氏(shi)(shi)定(ding)(ding)氮(dan)(dan)儀(yi)(yi),又名(ming)定(ding)(ding)氮(dan)(dan)儀(yi)(yi)、蛋白(bai)質測(ce)定(ding)(ding)儀(yi)(yi)、粗蛋白(bai)測(ce)定(ding)(ding)儀(yi)(yi)。
